Dr. Stephen Hursting discussed the strong links between obesity and cancer, highlighting evidence from major AICR/WCRF and IARC reports identifying 12–13 obesity-associated cancers. He explained that obesity promotes cancer through metabolic and inflammatory changes such as hyperinsulinemia, dysglycemia, altered adipokines, dyslipidemia, and immune suppression. In both human and animal studies, modest weight loss often was not enough to reverse these effects; reductions of roughly 10% or more appeared necessary to shift biomarkers and reduce tumor growth.<br /><br />He then reviewed interventions that may break the obesity-cancer connection, including bariatric surgery, calorie restriction, and especially GLP-1 receptor agonists. While human data on GLP-1 drugs remain limited, early epidemiologic studies are encouraging. His lab’s mouse studies with terzepatide showed substantial weight loss, improved metabolic markers, and reduced growth of breast, colon, and endometrial tumors. Mechanistically, the drug did not appear to act directly on cancer cells; instead, it reversed obesity-driven immune dysfunction, restoring CD8 T-cell anti-tumor activity and improving macrophage function.<br /><br />The talk concluded that GLP-1/GIP therapies may become scalable tools for cancer prevention and interception, but more human data, combination strategies, and long-term studies are still needed.
